Simone Zaza gave Sassuolo an early lead but Juventus hit back in the 35th minute when Carlos Tevez teamed with Claudio Marchisio with his deflected shot going past the home keeper.
Marchisio then put Juventus ahead in the second half and Spaniard Fernando Llorente sealed the three points with a back-heel from close range on 76 minutes.
“We have taken a very important step toward a historic third title,” Juve coach Antonio Conte said.
“We are having a super season. I hear talk about an out of this world Roma side but we are eight points ahead of them and in the Europa League semifinals. So find adjectives for us.”
Next week the black and whites could seal the deal at home to Atalanta with a win seeing them crowned Serie A champions.
Before that, Juventus hosts Benfica on Thursday in their second leg Europa League semi-final.
The Italians are 2-1 down from the first leg.
